El flavonoide cb2 obtenido de chromolaena bullata induce activación de células dendríticas humanas
Abstract
CB2, al igual que los otros flavonoides obtenidos de Chromolaena bullata, fue evaluado sobre células dendríticas y células tumorales humanas, para determinar su actividad antitumoral o inmunorreguladora. Sobre las células tumorales (A375 melanoma humano) induce una depolarización temprana de la membrana mitocondrial, la cual es reversible y no conduce a la muerte. En contraste, el tratamiento de células dendríticas humanas obtenidas a partir de monocitos de sangre periférica cultivados en presencia de factor de crecimiento de granulocitos y monocitos (GM -CSF), e interleucina 4 (IL-4) durante 5 dias; induce activación de estas células. Con el fin de descartar la presencia de Lipopolisacárido (LPS) en la preparación del flavonoide, el cual puede ser el causante de la activación, evaluamos la presencia de este LPS con el test del Lisado de amebocitos de Limulus (LAL) y solo se utilizaron las muestras con menos de menor 1.0 unidad de endotoxina por 20 ug de proteina. Con estas precauciones, observamos claramente que el CB2 aumenta la expresión de moléculas coestimulatorias como CD83 y CD86 necesarias en la presentación antigénica. La incubación de las células con el CB2 y Polimixina B, la cual captura el LPS no disminuye la expresión de estos marcadores, corroborando el papel activador de este flavonoide sobre las células dendríticas humanas.Downloads
